The quantitative separation of purines, pyrimidines, and nucleosides by paper chromatography

By paper strip chromatography in a butyl alcohol system, cytosine, uracil, adenine, and thymine may be isolated from mixtures and from hydrolysates of nucleic acids; the migrating substances, extracted from segments of the paper, are identified and their quantity detd. by u.-v. spectrophotome-try. Guanine also can be recovered under favorable circumstances. In the same way the nucleosides cytidine, guanosine, adenosine, and thymidine are separable from each other and from most of the free bases. The composition of binary mixtures, if encountered, may be detd. quantitatively. The method outlined appears to provide a means of estimating with some accuracy the constituent purine and pyrimidine bases, and probably the nucleosides, in hydrolysates of nucleic acids. Chromatography of partial hydrolysates of nucleic acids reveals the rate of liberation of individual bases and nucleosides.
