Transfer of charged and uncharged molecules in the placenta of the rabbit

The fetal placental circulation of the rabbit was artificially perfused with rabbit blood containing acetylene and labeled water, urea, chloride ions, and sodium ions. Maternal placental blood flow was calculated on the basis of acetylene losses from the perfusate. Equilibration periods of 20-30 min. were allowed after each change in the isotope content of the perfusate or in the rate of perfusion to ensure a steady state. The results were analyzed by means of a graphical method and also by a method of approximation that did not require assumptions regarding the vascular arrangement of the rabbit placenta. The 95% confidence limits of the mean permeability of the rabbit placenta at 27-29 days of gestation were: for water 3.9-8.5 ml/min., for sodium ions 0.07-0.15 ml/min., for chloride ions 0.14-0.28 ml/min., and for urea 0.26-0.42 ml/min. Positive or negative charges do not grossly affect the permeability of molecules in the rabbit placenta.
