Gomori's technic for the demonstration of alkaline phosphatase was applied to cultures of normal and (for the first time) to neoplastic tissues. In normal fibroblasts, alkaline phosphatase is located principally in the nucleus; weakly positive granules may be seen in the cytoplasm of cells undergoing degeneration. Fibroblasts of methylcholanthrene tumor cultures show marked phosphatase activity, constantly located in the nucleoli; weak reactions are given by nuclear substance. Cells of Roffo tumor cultures gave rather erratic nucleolar reactions but they had cytoplasmic granules which showed phosphatase activity. The authors discuss the relationship between phosphatase activity and cell growth and compare data obtained from normal and tumor cultures.
