Chronic alpha-tocopherol administration protects against behavioral impairments induced following intrahippocampal aggregated Abeta injections

Male Sprague-Dawley rats were trained under a Modified Recycling Conjunctive (MRC) schedule of reinforcement. Under this schedule subjects were exposed to repeating 30 second intervals over a 40 minute session. Lever presses during the first and last 10 seconds of each 30 second interval had no consequences. However, responses made during the middle 10 seconds resulted in the delivery of a reinforcer at the end of the 30 second interval. The function obtained by plotting the number of responses vs one second bins produced a bell curve with a peak of responding during the middle 10 seconds. When responding showed no trends subjects were bilaterally injected into the CA3 region of the hippocampus with 5 mul aggregated Abeta suspension. One group of subjects received 150 mg/kg alpha-tocopherol daily (orally) and the remainder received the vehicle in which alpha-tcopherol was suspended. Thirty days post injection the peak of the response function flattened for the Abeta injected group which did not receive alpha-tocopherol. This indicated a loss of temporal discrimination. Previous studies have shown that it takes approximately 30 days following Abeta injection before a behavioral detriment becomes apparent. alpha-Tocopherol may have brought about its protective effect through a reduction of oxidative stress following aggregated Abeta injection.
