Different Alpha Adrenoceptor Vasoconstrictor Mechanisms in Murine Mesenteric Arteries and Veins

We have shown that alpha-2 ((2) adrenoceptors (AR) do not mediate direct vasoconstriction of murine mesenteric arteries and veins. We tested for an indirect role of these receptors and of (1-AR in mesenteric arteries and veins from control and deoxycorticosterone acetate-salt (DOCA-salt) mice. Changes in vessel diameter were monitored in vitro by computer-assisted video microscopy. No differences were seen between control and DOCA-salt vessels. Constrictions to norepinephrine (NE) were competitively antagonized by the (1-AR antagonist prazosin (Praz; 3, 30, 300 nM) in arteries and veins. Yohimbine (Yoh; 3, 30, 300 nM), an (2-AR antagonist, competitively antagonized NE-induced constrictions in veins. Yoh did not affect the NE-elicited responses in arteries. Schild analysis revealed that in arteries but not veins, Praz acted as a competitive (1 antagonist (slope not different from 1). Similar analyses for Yoh in arteries revealed a non-linear relation in arteries but a linear relation (slope not different from 1) in veins, suggesting that (2 receptors contribute to the NE-induced vasoconstriction in veins. These data suggest the existence of a single population of (1 receptors in mesenteric arteries while in veins NE could be acting at (1 and (2 or multiple subtypes of (1 receptors. Altogether, these results suggest differences in adrenergic vasoconstrictive regulation between arteries and veins. .
