Differential behavioral effects of nicotine in the alpha7, beta2, and beta3 nicotinic receptor null mutant mice

We have initiated studies using the alpha7, beta2, and beta3 nicotinic receptor subunit null mutant mice to identify possible behavioral roles of these receptor subunits. Previous studies from our laboratory have demonstrated that acute nicotine injection produces changes in y-maze activity, open-field activity, body temperature, and acoustic startle in inbred mouse strains. These same behavioral measures were examined in the alpha7, beta2, and beta3 null mutant, heterozygous, and wildtype mice after acute i.p. injection with one of six doses of nicotine (0, 0.25, 0.50, 0.75, 1.0 and 1.5 mg/kg). The alpha7 and beta3 null mutants did not differ from their respective wildtype controls in y-maze activities, open-field activity, and nicotine-induced hypothermia for any of the doses of nicotine tested. However, the beta2 null mutants showed less sedation after 0.5 and 0.75 mg/kg nicotine in the y-maze and openfield compared to wildtype controls. The beta2 null mutant mice also showed attenuated nicotine-induced hypothermia at the 0.5, 0.75, and 1.0 mg/kg nicotine doses. The finding that beta2 null mutants differ in sensitivity to nicotine effects on some of the behavioral measures examined is consistent with previous studies from our laboratory that suggest that alpha4beta2-type receptors modulate these responses to nicotine. Thus far, a behavioral role for the alpha7 and beta3 subunits has not been found for behaviors we have examined.
