Effects of uridine 5-monophosphate on body weight, feeding and expression of neuropeptide Y in the paraventricular nucleus in male rats

The present study was aimed to evaluate the impact of chronic s.c. injection of uridine monophohate (5'-UMP) on daily feeding pattern, body weight and NPY expression. Male Sprague-Dawley rats (n=24, bw 200-250g) were separately raised in individual plastic boxes with food and water ad libitum. Animals were administered s.c. once daily with 5'-UMP (50, 100 or 200 mg/kg, n=6) or saline vehicle 30 min before lights off. Food intake was measured every 2 hours after lights off until lights on for 7 days. NPY levels of paraventricular nucleus (PVN) detected by immunocytochemistry was semi-quantitated using a computerized image analysis system. Average of body weight gains was significantly increased after 7-day injection of 100 mg/Kg 5'-UMP when compared with saline group. A slight increase of food intake was also observed following administration of 5'-UMP (F=2.46, p=0.09). 5'-UMP treatment also significantly increased the expression for NPY in the PVN of hypothalamus.
