A multicenter evaluation was conducted to compare the performance of moxifloxacin (MXF) and gatifloxacin (GAT) on a MicroScan Dried Overnight panel (TEST) with an NCCLS broth microdilution reference (REF) panel. The clinical study was conducted at three sites and consisted of three phases: efficacy (EFF), reproducibility (REP), and challenge (CHA). A total of 1035 clinical isolates, comprised of 340 gram-negative (GN) and 695 gram-positive (GP) organisms, were tested during the EFF phase. Twenty-five GN and 25 GP organisms, which included the recommended NCCLS Quality Control strains and isolates which generated on-scale MICs, were tested during the REP phase at all sites. During the CHA phase, 75 GN and 75 GP isolates were evaluated at one site. Essential agreement in the EFF phase between the TEST and REF panels for the GN isolates was 99.4% for both MXF and GAT (338/340) and for the GP isolates was 98.6% (685/695) for MXF and 98.4% (684/695) for GAT. Essential agreement for the CHA phase was 98.7% (74/75) with GN isolates and 100% with GP isolates for both MXF and GAT. The interlaboratory REP was 98.7% (74/75) for GAT with GN isolates and 100% with GP isolates. REP for MXF was 97.3% (73/75) with GN isolates and 98.6% (73/74) for GP isolates. The performance of moxifloxacin and gatifloxacin on MicroScan Dried Overnight panels gave excellent correlation in comparison to an NCCLS broth microdilution reference panels.
