To evaluate how basal ganglia have access to different frontal motor areas, multiple anterograde and retrograde tracers were injected in the cortex, pallidum and thalamus in macaque monkeys. The distributions of labeled cells and terminals were related to thalamic parcellation and basal ganglia compartmentalization based on multiple architectonic criteria. Areas pre-SMA and SMA share common input from pallidal afferented thalamic ventral anterior (VA), ventral lateral anterior (VLa), dorsal part of the ventral lateral posterior (VLpd) and ventral medial (VM) nuclei. However, there is a rostral to caudal gradient of pallidal inputs to thalamocortical projections to pre-SMA, through VA, and to SMA, through VLa. Pre-SMA, in addition, receives strong input from the mediodorsal (MD) nucleus and from non-pallidal area X which have more minor or no projections to SMA. Inputs to PMd originate from the same thalamic territories projecting to pre-SMA and/or SMA (e.g. VA, VLa, VLpd, area X), but in addition, from the ventral part of the ventral lateral posterior (VLpv) nucleus which receives only minor pallidal input and projects massively to M1. In the medial thalamus, there is evidence for particularly more extensive projections from the central lateral (CL) nucleus to pre-SMA than to the other motor cortical areas. These results provide additional evidence for a differentiated influence of basal ganglia onto distinct frontal cortical areas through pallidothalamocortical pathways.
