Treatment effects of DHEA-sulphate on adiposity and substrate metabolism in female swine

The metabolic effect of DS-treatment (tx) with and without the aromatase inhibitor fadrozole (FAD) on steroid hormones and substrate disappearance (dis), as measured by indirect calorimety, was investigated. Prepubertal miniature pigs (n=24), isocalorically-fed, were treated continuously for 4 wks with either DS, FAD, DS+FAD, or control (C). DS-tx raised the ratios of plasma DS:testosterone (T) (p<0.01), DHEA:T (p<0.01), and decreased log T levels (p<0.02). FAD-tx reduced DS:T ratio (p<0.01). An increase in backfat measured by ultrasonography (p=0.06) was associated with a decrease in the rate of lipid dis (p<0.01) in all DS-tx animals. Plasma insulin levels of DS-tx swine were increased (p<0.02) vs C; there were no differences in glucose levels. Lipid dis of all swine was positively associated with T levels (p<0.01) and negatively with both DS:T (p<0.05) and DHEA:T ratios (p<0.02). Plasma DHEA levels were positively associated with triglycerides (p<0.01). Collectively, findings indicate imbalances in steroid hormones associated with DS-tx produce a decrease in lipid disappearance and an increase in fat storage in prepubertal female swine.
