VEGF, VEGF isoforms and VEGFR2 in human skeletal muscle

The VEGF isoforms and their receptors are prime regulators, with overlapping but specific roles, in controlling the neovascularization. In the present study the main aim was to explore at the mRNA level if exercise induces a concurrent increase in VEGF and VEGF receptors and whether exercise induces an increase in specific VEGF isoforms. Additionally, we wanted to study if there was an additive response following exercise performed in a condition of enhanced metabolic stress.Method: Nine healthy male subjects performed a 45'bout of one-legged exercise during reduced blood flow as well as under normal flow condition. Biopsies were taken in vastus lateralis before and 2h and 6h after exercise. mRNA levels were analyzed with real time PCR.Results: Total VEGF and the isoforms 121,165 and 189 were all increased at 2h after exercise but with no further increase at 6h. The VEGFR2 was increased at 6h after exercise. For all measured factors, the mRNA increase was greater after exercise with reduced blood flow compared to exercise under normal blood flow condition.The present data with an increase in both the VEGF and VEGFR2 mRNA supports the hypothesis that the VEGF-system is involved in exercise-induced angiogenesis. Furthermore, an increase of all the major VEGF isoforms occurs in response to exercise. Additionally, the greater mRNA increase in response to exercise with reduced blood flow indicate ischemia and/or related metabolic events as possible regulating stimuli.
