Microchromatography for determination of hemoglobin A 2: statistical study of values in normal subjects and beta-thalassemic heterozygotic subjects

An improved simplified method for identifying abnormal hemoglobin (HbA2) in heterozygotic .beta.-thalassemia was reported. It is a microchromatographic method, using DE-52. The normal values (of control persons) and abnormal values for HbA2 are limited to a small range and completely separated from those with the genetically determined hemolytic anemia. Accuracy of the method was demonstrated in 2 normals and 1 thalassemic individual. Twelve tests were done on each individual. Average values and standard deviations were reported. In 9 cases of sideropenic hypochromic anemia the average value of HbA2 was 1.61%, with S.D. = 0.150. In the thalassemic case the average value for HbA2 was 4.60 .+-. 0.34 with S.D. = 0.389. The method was adaptable to determining Hb S and Hb D-Punjab, and for Hb C.
