Mouse erythroleukemia cells were used for experimental work; they resemble proerythroblasts and basophilic erythroblasts morphologically, exhibit several other phenotypic markers of erythroid cells and can be maintained in suspension culture. Determination of the primary sequences of the various globin mRNA's provided the ultimate proof for selective codon use and, therefore, for the selective requirements for various isoaccepting tRNA's in animal cells. The structural basis for many hemoglobinopathies were also defined. The mRNA sequences reveal sites where gene rearrangements might occur and provide the necessary information to make initial predictions regarding secondary structure and interaction of mRNA with the ribosomes. The molecular lesions in thalassemia were intensely studied to provide insight into the mechanisms of gene regulation.
