Treatment with 5-fluorouracil: randomized study comparing an oral preparation with intravenous administration

Patients (94) with various neoplasms were randomly treated with i.v. or oral (p.o.) fluorouracil (FU). In the 1st phase (46 patients), patients were treated with equal doses of FU in the 2 treatment groups; in the 2nd phase (48 patients), a 30-40% larger dose of FU was administered in the p.o. group. In neither phase were substantial differences in efficacy of FU administered i.v. or p.o. observed. As to toxicity, in both phases some increase in nausea and vomiting was observed using oral FU; only in the 2nd phase a greater incidence of leuko-thrombocytopenia was observed in the p.o. group.
