Evaluation of TNF-alpha, IL-8 and IL-10 transcriptional activity in milk from healthy dairy cows during lactation period

This study was conducted to measure the levels of normal transcriptional activity of 3 important cytokines (tumour necrosis factor alpha , interleukin 8 and interleukin 10) in milk from healthy cows and to determine the lactation stage-dependent changes in levels of these mediators. 50 second- and third-lactation healthy cows were grouped according to 3 different stages during the lactation cycle: early lactation (n=12); midlactation (n=18) and late lactation (n=20). Samples from early and late lactation periods tended to express lower levels of TNF- alpha and IL-8 than those from midlactation period. The IL-10 expression level in samples from early- and late-lactation tended to be higher than those from midlactation. The levels of cytokines in the 3 lactation groups were insignificantly different. Results showed that there were significant differences in the total profile of the 3 cytokines over the different lactation periods. Significant correlations both between IL-8 and IL-10 in midlactation period and among the 3 cytokines during late-lactation were observed. Cytokine transcription levels were more correlated in the late-lactation as compared with early- and midlactation periods.
