Abstract
A fast and simple radioimmunoassay (RIA) technique was developed for an antibody to DNA .times. RNA hybrid using protein A-bearing Staphylococcus aureus cells as immunoadsorbent and a glass microfiber filter for the separation of free antigen and antibody-antigen complex. A simple method for preparing 3H-labeled DNA .cntdot. RNA hybrid using single-stranded circular DNA of viruses and Escherichia coli RNA polymerase (nucleosidetriphosphate: RNA nucleotidyltransferase, EC 2.7.7.6) is also described. In comparison to a hybrid made of natural sequences, a synthetic homopolymer hybrid poly(A) .cntdot. poly(dT) was a poor competitor for the antibody by this RIA technique.
